Abstract:
Shoot tip explants of Spiraea were cultured in vitro on Linsmaier and Skoog medium gelled with agar and containing benzyladenine at pH 5.6. The explants were cultured for one month prior to placement in uncapped jars in a sealed glass tank.
A tube with a millipore filter was passed through a small hole in each tank and was used to supply gases to the tank.
No stimulation of shoot proliferation (shoot number) occurred in treatments with metabolic gases when compared with the control tank (no gas flow). The mean shoot numbers were 1) control - 83, 2) oxygen - 86, 3) air - 89. However, shoot number in the nonmetabolic gas (N2) treatment increased significantly over all other treatments - 4) nitrogen - 115 (p<0.05).
