Abstract:
In tissue culture, contamination can be controlled by early detection and by prevention of spread.
Both factors have been studied in cultures of Malus.
Contamination of plants was detected at an early stage by culturing on media which promote bacterial growth.
Best detection was achieved when the conditions for bacterial growth were optimal.
The spread of bacterial contamination was caused by insufficient flaming of contaminated tools (1 to 3 sec) and by survival of bacteria in 96% ethanol (for at least 40 min). Addition of Burtan, Dettol and NaOCl to the ethanol reduced bacterial survival only partly.
Flaming for 5 sec or more did eliminate the spread of bacterial contamination at transfers.
