SPECIFIC IDENTIFICATION OF ERWINIA AMYLOVORA THROUGH THEIR METABOLITES

Abstract:
Use has been made as antigen in ELISA of diluted supernatants obtained after centrifugation of liquid culture media inoculated with E. amylovora and other bacterial species.

After 24 hours of incubation of 64 strains of E. amylovora, the corrected optical density (cOD) observed reached a mean value of 1.250 when the supernatants were diluted 100 times. After 48 hours of incubation, the cOD mean values were respectively 1.600 and 1.345 for 100 and 500 times diluted supernatants.

In contrast, the cOD mean value obtained for 90 strains of other bacterial species was lower than 0.100 after 24 and after 48 hours of incubation.

It has also been shown that the association of E. herbicola, or P. fluorescens or P. syringae with E. amylovora in a same culture medium did not disturb the detection of this latter. The cOD values observed after 24 hours of incubation of mixed cultures were at least 2.5 times higher than those obtained for unmixed bacterial cultures (supernatant diluted 100 times) and after 48 hours, respectively 2.6 and 1.5 higher for 100 and 500 diluted supernatants of the mixed cultures.

The use in ELISA of metabolites produced by E. amylovora allows thus a good and specific identification of this bacterium.

Results of the first experiments with plant material are reported and discussed.
