VEGETATIVE PROPAGATION OF ANDROGENOUS EMBRYOS OF HORSE CHESTNUT BY MERISTEM CULTURE IN VITRO

Abstract:
Micropropagation of several clones by meristem tip cultures from haploid embryos, dormant buds of young or adult Aesculus hippocastanum L. trees and their sprouts was achieved. For successful meristem differentiation into leaf rosettes, two mineral solutions were used: a. solution containing macro- and microelements after Quoirin and Lepoivre ( 1977 ) and b. solution containing half-strength macroelements of Murashige and Skoog ( 1962 ) and microelements after Nitsch and Nitsch ( 1969 ). Both solutions contained 2% sucrose, 0.7% agar, vitamin mixture after Jacquiot ( Gautheret, 1958 ) and hormones ( in mg· 1-1 ): 2,4-D 0.001, BAP 1.0 and GA3 0.1. Axillary branching of the rosettes was obtained in solution a enriched by vitamin mixture after Walkey ( 1969 ) and hormones ( in mg· l-1 ) : BAP 1.0 and GA3 0.1. Shoot elongation was induced in the same medium enriched with vitamins after Jacquiot ( Gautheret, 1958 ) and 1.0–10.0 mg· 1-1 of GA3. Number of elongated shoots was rather low. Rooting was achieved in solution a, where macroelements were reduced to half strength and which was supplemented with vitamin mixture after Walkey ( 1969 ) and 2.0 mg· l-1 of IBA. Cultivation in the dark was beneficial for rooting of the shoots of some Aesculus clones originating either from haploid or diploid plants.
