IN VITRO PROPAGATION OF ADULT MATERIAL AND SEEDLINGS OF CORYLUS AVELLANA

Abstract:
Adult and seedling explants of Corylus avellana L. cv. Gironell were cultured in vitro on different cytokinins supplemented media. The cytokinins tested were Zeatin, BA, 2 i P and Kinetin at concentrations ranging from 0 to 20 mg/l. The basal medium was the MS formulation at half strengh supplemented with 0, 01 mg/l AIA, 2 mg/l GA, 30000 mg/l saccharose and 8000 mg/l agar. Shoot segments about 2–3 cm long with one vegetative bud were used as explants. Cytokinins tested promote hazelnut budding at dose between 2,5 – 10 mg/l. Adult explants had given less multiplication rate that juvenile ones at all concentration tested but with similar behaviour patterns. This suggest that the micropropagation is not only a problem concerning with chemical composition of culture media but that hazelnut presents juvenility-agening problems similar to other woody plant species, and it would be necessary to submit adult material to a rejuvenation process simultaneusly to optimizise culture medium.
