MICROPROPAGATION OF PRUNUS TENELLA CV. FIREHILL

Abstract:
Shoot cultures of Prunus tenella cv. Firehill have been initiated from shoot tips taken from containerised stockplants in June and cultured on Murashige and Skoog medium (M & S) containing benzylaminopurine (BAP). In the presence of 1 mg 1-1 BAP' proliferation of axillary shoots was enhanced but, in many cultures, necrosis of shoot tips and vitrification occurred. Vitrification of shoots appeared to be stimulated by stress to the explants during surface sterilisation and by a high concentration of BAP in the culture medium. Vitrification could not be reversed by subculturing shoots onto media containing <1.0 mg 1-1 BAP. Rooting of cultured shoots was achieved with or without auxin in the medium. Swollen roots and callusing were associated with naphthaleneacetic acid (NAA) whereas normal thin roots were produced on media lacking auxin or containing indolebutyric acid (IBA).
