IN VITRO PROBLEMS RELATED TO PROPAGATION OF DIFFERENT FERN SPECIES

Abstract:
Gametophyte and callus cultures were initiated from surface sterilized spores of different fern species and have been maintained on Moore's or/and MS media containing 0.5% saccharose. The effect of various auxins (IAA, NAA) and cytokinins (kinetin, BA) was investigated. Cultures were grown in light.

Apogamous sporophyte organisation has been observed in the species Pteridium aquilinum, Cyclosorus contiguus, Dryopteris filixmas, Pteris henryi, Osmunda regalis and recently in Pteris cretica cv. albolineata. Cytological test proved their haploid level.

Sporophytes of Marsilea quadrifolia and Pilularia globulifera could be also regenerated from excised shoots.

Four different callus types were observed and investigated.

Sporophytes are maintained on half-strength (MS/2) hormon free media and then transferred to soil.
