ISHS Acta Horticulturae 212: Symposium on In Vitro Problems Related to Mass Propagation of Horticultural Plants

ROOTING AND ACCLIMATIZATION OF PELARGONIUM ZONALE PLANTLETS

Author:   A. Aldrufeu
Abstract:
The problem of non-functional roots often arises in the process of rooting in vitro in agar. To avoid some of the negative effects that the rooting in vitro presents, we have studied the effect of different substrates as supports for the culture medium.

Plantlets of Pelargonium zonale var. Rubin multiplied in vitro are transferred to a rooting medium with and without saccharose, using as support: agar, cellulose, sand, perlite, vermiculite, Floratorf peat and TKS-1 peat.

In most media the percentage of rooted plants is higher than 90%.

The addition of 20 g/l of sucrose to the medium increased the quantity of rooted plants, and the number of roots per plant. Whereas the length of the roots depended on the substrate used.

All plants alive were transferred in vivo to the same substrate -1 vermiculite: 1 TKS-1 peat-. Those showing the best results came from perlite (> 95%) independent of whether they had saccharose or not.
