ETHYLENEACCUMULATION IN CULTURE VESSELS - A REASON FOR VITRIFICATION?

Abstract:
In tissue-cultured Pediocactus species vitrification can only be overcome by high agar concentration, reduced humidity and good gas exchange. Lowering the total water potential seems to be the most effective way to prevent vitrification. Decreased NH4NO3 and kinetin concentration had no effect on vitrification. Vitrified shoots could be normalized by grafting them. Measurements of ethylene. (C2H4) showed that different brands of agar discharged much C2H4 when they were exposed to the light, but not in the dark.

Charcoal (CH) was able to absorb the C2H4 released by the medium, and in addition the C2H4 produced by explants.
