EFFECT OF HEAT TREATMENT ON ACCELERATION CHRYSANTHEMUM MULTIPLICATION BY MERISTEM-TIP CULTURE

Abstract:
In an effort to improve the efficiency of the meristem culture method, plants were subjected to a temperature of 37 °C for 8 weeks, prior to isolation of the meristems. Cultivated varieties were obtained from meristem-tips of lateral shoots, excised at the length of 0.4–0.5 mm. They were planted on Murashige Skoog culture medium, changing the concentration of different components. That variety of medium proved to be the best which contained Heller-4 modified trace element solution. Meristem-tips from heat treated plants grew more rapidly and survived better, whereas 93 % of meristem-tips started to proliferate in comparison with 83.6 % of meristem-tips from non heat treated plants. Throughout the culture media used for multiplication cultivated varieties, the best results were observed with the shoot tips taken from heat treated plants which grew more rapidly (high rate of multiplication, high average number of adventitious shoots, height and number of leaves) than those grown from untreated plants. In stimulating media for root formation, the shoot tips taken from heat treated plants, the root formation started very early. The plantlets having roots were tested in glass house planted in the mixture of peat moss and perlit (3/1 or v/v) through 4 weeks.
