DEVELOPMENT OF A SIMPLE METHOD FOR COLLECTING AND PROPAGATING COCOA GERMPLASM IN VITRO

Abstract:
Efforts to collect, conserve and distribute the germplasm of cocoa are hindered by the deterioration of samples in transit. A simple in vitro culture based method has been developed to maintain viability in such material for sufficient time for it to be transported safely from the field to genebanks and to users. Explants are treated with a benigh surface sterilant and inoculated into a semi-solid medium containing antimicrobial agents. A number of compounds have been screened for their efficacy in controlling contamination. FBC Protectant Fungicide and drinking water purifying tablets are effective surface sterilants, and the fungicide Tilt MBC and the antibiotics rifamycin and trimethopuim are effective culture medium additives. Some limited spontaneous rooting has been observed and collected material has successfully been grafted onto seedlings.
