BACTERIA CONTAMINANTS IN SHOOT CULTURES OF PRUNUS AVIUM L. CHOICE AND PHYTOTOXICITY OF ANTIBIOTICS

Abstract:
In the process of in vitro propagation of wild cherry tree (Prunus avium L.), bacterial contamination may occur after several transfers and could result in the loss of clones.

In contaminated material, isolated bacteria belong to different genera. To eliminated these contaminants it is necessary to know effect of antibiotics on bacteria and also on explants. Aminoglycosides, tetracyclines and Rifampicin have the broadest spectrum of activity against contaminants. Non contaminated explants were treated with 4 concentrations of antibiotics during 7 days in agitated liquid medium. After Tetracyclin treatment there is no loss of material, but aminoglycosides induce chlorosis, necrosis and death of explant at high concentration. There is a great variability between clones in susceptibility to all antibiotics.

With this method five contaminated clones were treated. After four transfers three are still healthy.
