Abstract:
In vitro shoot-tip micrografting has been developed to recover virus-free Prunus plants.
Shoot meristem apices, excised from two species of Prunus cultivars, at the juvenile and adult stages, were grafted in vitro on to disease free rootstocks.
About 50% of the grafts lived with juvenile Prunus apices, but only 15% to 20% success was obtained with adult Prunus meristem grafting.
The survival rate of the tips was directly related to their size.
The behaviour of these two materials after micrografting was compared.
The grafting problems with adult material, the effect of numerous parameters (physiological status of plants, medium conditions…) and the incidence of virus-free plants are discussed.
