NUTRITIONAL ENVIRONMENT AND REDIFFERENTIATION FROM CULTURED TISSUES OF AZADIRACHTA

Abstract:
The callus raised from the young stem bark of Azadirachta indica A. Juss. and maintained for one and a half years was completely devoid of nimbin, an important secondary metabolite (triterpene). Redifferentiation of compounds and organs (and plantlet production) by proper nutritional balance, were the object of the paper. After several trials with inorganic salts of Murashige and Skoog medium supplemented with various auxins, cytokinins, vitamins and natural supplements for organogenesis and plantlet formation, we found high benzylaminopurine and low napthalene acetic acid produced good shootbuds while the reverse induced roots. Indole butyric acid (0.5 mg/l) when added to the best shoot producing combination of ingredients, produced shoot and root together. Nimbin synthesis increased markedly, only with organogenesis. Root formation (alone) accompanied the least quantity of nimbin, shoot alone accompanied more nimbin synthesis, differentiation of root and shoot together accompanied the highest nimbin synthesis but less than in cotyledons.
