THE DEVELOPMENT OF ELISA TO DETERMINE ABSCISIC ACID AND INDOLE-3-ACETIC ACID IN BULBOUS CROPS

Authors:   J.M. Franssen, B.E. Snaar-Jagalska, C.Th.C. van der Hulst
Abstract:
A competitive ELISA was developed for the determination of the endogenous quantity of abscisic acid (ABA) and indole-3-acetic acid (IAA) in bulbous crops. Therefore, ABA and IAA were conjugated to bovine serum albumin by their carboxyl groups. Antibodies were raised in rabbits. With these antisera a good, reproducible, standard curve for ABA was developed. The standard curve for IAA, however, is less sensitive.

It was not possible to determine IAA or ABA levels in crude plant extracts. To determine ABA levels in flower-buds of lily several purification steps were tried and will be presented, but the results are still rather variable.
