Abstract:
An experiment was carried out to determine the auxin requirement during the course of rooting of shoots from shoot tip culture of Prunus cerasus cv. 'Schattenmorelle'. There was no difference in mean number of roots per shoot whether shoots have been subcultured for 22 days on auxin (1 ppm indolebutyric acid) containing medium or transferred after 3 respectively 8 days to auxin free medium.
The mean number of roots per shoot (x= 4.1) was maximal when shoots have been immersed for 3 sec. in a aqueous solution of 200 ppm IBA before subculturing on auxin free medium.
In this case roots emerged earliest.
A comparison of root length growth rates on auxin free and auxin containing media revealed no requirement of exogenous auxin for root elongation.
Isoperoxidase number and activity of stems have been comparatively analysed by isoelectric focusing.
Isoperoxidase pattern did not reflect the quantitative differences in rooting within different treatments.
Isoperoxidase pattern developed according to the chronological stage of stem growth but auxin treatment did not change the variety specific pattern.
