IN VITRO CULTURES OF MERISTEM TIPS OF CANNA INDICA /L./

Abstract:
The investigations performed were intended to develop a method for propagation of Canna indica from meristem culture in order to obtain plants free of viruses. To this end the authors used agar solidified and liquid MS medium with the addition of growth regulators and enriched with ascorbic acid, malt and yeast extracts. The cut out meristems developed better in liquid /½ MS/ than in agar solidified medium. Auxins, cytokinins and special additions present in the nutrient solution ensured a better survival of explants and influenced their growth and development. Buds developed from the isolated meristems, obtained in experiments after a 6 month oulture were kept at temperature of 6°C for four winter months. In early spring the passaged buds quickly developed shoots and roots. Nutrient solution with the addition of kinetin /2 mg/dm3/, adenine sulfate /100 mg/dm3/ and NAA /0.2 mg/dm3/ appeared to be particularly advantageous in this respect. The plants grown in such medium were propagated by axillary buds, cut off and transferred to fresh nutrient solution, started to grow in a short time. In this way a great number of Canna plants may be obtained in vitro conditions.
