EFFECTS OF BA, RHIZOME LENGTH, MECHANICAL TREATMENT AND LIQUID SHAKING CULTURE ON THE SHOOT FORMATION FROM RHIZOME IN CYMBIDIUM FABERI ROLFE ROLFE

Abstract:
The rhizome tips derived from cross breeding of 'Ro-Shanghai-bai' with 'Kin-o-so' of Cymbidium faberi Rolfe were cultured in the modified Kyoto solution supplemented with N6-Benzyladenine (BA). The rate of shoot formation per rhizome was not affected by the length of inoculated rhizome but affected greatly by BA concentration. No shoot was formed without BA. The rate of shoot formation was 90% at 0.1 ppm BA and about 100% at 1 and 10 ppm BA. The number of shoots per rhizome was affected greater by BA concentration than rhizome length. The shoot growth was prevented with 10 ppm BA. The mechanical treatments such as the destruction of rhizome meristem by the resection of rhizome apex or the pricking with a needle and the vertical halving of node or internode resulted in the augmentation of death of explants. For shoot formation, BA affected the mechanically treated and surviving explants similarly as the non-treated explants. In liquid shaking culture, the organ formation and shoot growth on the rhizome were abnormal and terminal buds and axillary ones grew hypertrophicaly at high concentration of BA. After transplantation of the mass of underdeveloped rhizomes to the agar medium without BA, many of them developed into new rhizomes and some of them formed callus-like rhizomes.
