Abstract:
Complementary DNA (cDNA) was prepared by oligo-dT priming of RNA of a gladiolus isolate of bean yellow mosaic virus (BYMV). The cDNA was found to hybridize to RNA of several isolates of BYMV and of several other potyviruses bound to nitrocellulose.
Hybridization to nucleic acids in healthy plant tissue, or purified cucumber mosaic and tomato ring spot viruses, was negligible.
The cDNA hybridized to some extent with the RNAs of clover yellow vein virus (CYVV), iris mild mosaic virus (IMMV) and iris severe mosaic virus (ISMV)despite the fact that the coat protein of only CYVV is closely related to BYMV. IMMV and ISMV coat proteins do not react with BYMV coat protein antiserum in indirect ELISA, and are only distantly, if at all, serologically related to BYMV. The hybridization of these RNAs with a cDNA probe prepared from the 3' end of BYMV RNA is thus surprising, as the coat protein gene of the potyviruses is postulated to be the 3'-terminal segment of the genome.
