INACTIVATION OF VIROIDS IN CHRYSANTHEMUM BY LOW-TEMPERATURE TREATMENT AND MERISTEM-TIP CULTURE

Author:   N. Paludan
Keywords:   Chrysanthemum, chrysanthemum stunt and chrysanthemum chlorotic mottle viroid, inactivation, low-temperature treatment, meristem culture
Abstract:
Production of chrysanthemum free of chrysanthemum stunt viroid (CSV) and chrysanthemum chlorotic mottle viroid (CCMV) has so far been very difficult, and the results obtained have not been easy to reproduce.

Regarding inactivation, however, low-temperature treatment of some viroids has turned out to be just as effective as high-temperature treatment for viruses.

Experiments carried out at the Research centre with the CSV and CCMV have shown that both viroids became inactivated increasingly with the prolonged low-temperature treatment at 5°C combined with meristem-tip culture. The inactivation percentage ranged from 50 to 70.
