ISHS Acta Horticulturae 131: In Vitro Culture, XXI IHC

TOWARDS THE PROPAGATION OF TULIP IN VITRO

Authors:   P.G. Alderson, R.D. Rice, N.A. Wright
Abstract:
Reliable regeneration of adventitious shoots on explants of immature floral stems from tulip dry bulbs has been achieved, and was found to be influenced by the stage of development of the bulb, the thickness of the explant and the origin of the explant within the floral stem. Shoots 12 weeks of age contained developing meristematic centres at their base, and bulb formation was induced following cold treatment or exposure to gibberellins. Bulbing consistently occurred in shoots produced on floral stem explants grown at 20°C for 14 to 18 weeks followed by 4°C for 8 weeks and further incubation at 20°C. Shoots exposed to gibberellins in the culture medium, or as a soak in a sterile solution, also exhibited a bulbing response.
