As the basic units of organism, single living cells are the foundation of living activity and evolution.
The identification, separation and analysis of single living cells can be used to analyze deep level of heterogeneity and operation mechanism of a living system.
The study of fruit development is based on the extraction of active single cells.
By using the matured Fuji (Malus × domestica Borkh. 'Fuji') apples from the teaching and practicing base as the test material, this paper studies the impact of different enzyme liquid combinations and enzymolysis time on the single cell in apple flesh.
By using VBL to test cell wall, FDA to detect cell activity and fluo-4AM to determinate Ca2+ concentration, the results show that treating the apple flesh in 0.1% macerozyme solution for half an hour can provide a better condition for separating single cells.
