Genetic diversity analysis of Iranian melon (Cucumis melo L.) accessions using ISSR markers

Abstract:
Melon (Cucumis melo L.) is one of the old vegetables in Iran which is one of the centers of genetic diversity of melon. In order to study genetic diversity among and within 27 melon accessions, ISSR markers have been used. A total of 23 ISSR primers were screened, out of these 10 ISSR primers were selected based on degree of polymorphism. To appraise genetic population structure, GenAlex software was applied. Polymorphic information content and marker index were estimated to find distinction power of markers. (AG)8T primer had the highest for these yardsticks. Polymorphism percent mean for all of primers was 51.6%. Molecular variation analysis used for calculating percent share of genetic diversity among and within accessions in which 31% of total variation related to the variation of among accessions and 69% of that related to within accessions. The UPGMA tree based on Nei's unbiased genetic distance separated 27 populations into two main groups. ISSR markers could separate non-sweet melon group (dudaim) from sweet groups and placed accessions collected from same regions in same groups. In Iran, as one of the main centers of diversity, it is necessary to do more research to discover new and susceptible genotypes and retain them toapplying in breeding programs.
