Strawberry (Fragaria × ananassa) cultivars are susceptible to fungal diseases and their resistance is one of the most important desired economic traits for growers.
Use of genetic resources, carrying resistance genes, play an important role in breeding programs leading to resistant cultivars, which can have positive impact on environment and economy.
Therefore, the identification of resistance genes in modern strawberry cultivars and breeding materials, and then selection of the best resistance genes combination(s) are the first steps for a successful breeding.
Conventional plant breeding takes time and depends on ambient conditions.
The breeders are very interested in new technologies, such as molecular markers, that could make this procedure more efficient and faster.
In the present work, six strawberry cultivars: 'Mira', 'Idea', 'Benton', 'Marmolada', 'Elsanta' and 'Cambridge Favourite' were evaluated to determine the presence of resistant alleles Rpf1 to red stele root rot (Phytophthora fragariae) and Rca2 to anthracnose (Colletotrichum acutatum) using molecular markers: OPO-16C_438, SCAR-R1A_285 and STS-Rca2_240. The molecular assays demonstrated the presence of the resistant Rpf1 allele to red stele root rot in homozygous status for 'Benton' and 'Mira' and in heterozygous status for 'Idea'. For 'Cambridge Favourite', 'Marmolada' and 'Elsanta' the susceptible allele, rpf1 was detected.
Regarding Rca2 gene, molecular analysis showed that only 'Benton' carries the resistance allele, Rca2, but we cannot say if this gene is in homozygous or heterozygous status.
As a consequence, it can ascertain that 'Benton' contains the Rpf1 (RR) resistance gene to red stele root rot and Rca2 (RR or Rr) resistant gene to anthracnose.
Therefore, 'Benton' could be used for transfer of these genes or pyramiding more genes for resistance to red stele root rot and to anthracnose using markers assisted selection (MAS) in the strawberry breeding program.
