DNA polymorphism of two -galactosidase genes (-gal1 and -gal2) in Thai local and commercial cultivars

Abstract:
Papaya (Carica papaya L.) is a fast growing fruit crop with versatility for fresh consumption and for manufacturing into several food related and pharmaceutical and cosmetics products. Although papaya fruit with firm texture is preferred over the soften one, many Thai papaya cultivars display the rapid softening during fruit ripening. In this study, two β-galactosidase (β-gal1 and β-gal2) encoding degradation enzymes were amplified from 29 samples and the genetic variation was determined using denaturing polyacrylamide gel electrophoresis and sequence analysis. Of 18 exons and 17 introns in β-gal1, a 16 base pair insertion/deletion was found in intron 7 concurrently with 4 SNPs found in intron 7 and 8 in six papaya cultivars. In β-gal2, on the other hand, there is only one insertion/deletion of T was found in intron 11 of solo type papaya including 'Sunset Solo', 'Huangold' and 'Pluk Mai Lai'.
