ISHS Acta Horticulturae 110: V International Symposium on Virus diseases of Ornamental Plants

LATENT INFECTION OF IMPATIENS BY A POTEXVIRUS

Authors:   B.E.L. Lockhart, J. Betzold
Abstract:
During indexing of Impatiens for infection by tobacco ringspot and tobacco streak viruses, many of the plant samples were found to contain viruslike particles when PTA-stained leaf dip preparations were examined in the electron microscope. The virus was transmissible to healthy Impatiens, in which it appeared to cause no symptoms. In host range tests the virus produced local lesions without systemic invasion in Gomphrena globosa and Tetragonia expansa; symptomless systemic infection in Beta vulgaris; and local lesions and systemic mottle in Chenopodium album, C. amaranticolor and C. quinoa. No infection occurred on any of sixteen other indicator hosts including Phaseolus vulgaris, Vigna unguiculata, Cucumis sativus, Nicotiana clevelandii, Antirrhinum majus, Melilotus alba and Trifolium incarnatum. In crude leaf tissue extracts the virus had a particle length of 520 nm, a thermal inactivation point (TIP) of 68–72 °C, and remained infectious for at least 2 weeks at 22–24 °C. The virus was not transmitted in a nonpersistent manner by Myzus persicae, and did not react with antisera to any of six other potexviruses.
