Abstract:
The Derrick method of trapping increased numbers of virus particles on electron microscope support films is very sensitive when working well but has failed with certain viruses and under certain conditions.
Examples are given.
The protein A modification of the Derrick method was used with a small isometric virus (ryegrass cryptic virus) and a closterovirus (grapevine stem pitting-associated virus). Protein A gave up to a twofold but not a threefold increase in numbers of particles trapped, compared with the Derrick method alone, in contrast to the much greater increases claimed in published work.
Reasons for these results are suggested.
