DETECTION OF LILY SYMPTOMLESS VIRUS (LSV) IN BULB TISSUE OF LILY BY MEANS OF ELISA

Abstract:
Lily symptomless virus (LSV) was readily detected in bulb extracts of lily cv Enchantment by the routine ELISA procedure. For other cultivars, e.g. Destiny, Uchida, Grand Commander, Connecticut King, Uncle Sam and Brabander, this procedure had to be changed. To detect LSV reliably in bulb extracts of these cultivars, it was necessary to homogenize the tissue in an extraction buffer (pH 7.4) of 0.04 M phosphate and to incubate the extract with the enzyme hemicellulase (0.25 U/ml) for about 13 h at room temperature.
